Yes, you will need to freeze water in the ice packs provided in your kit, unless you have a Chiller unit which will cool the water for you.  

Note: the use of a chiller unit will support in maintaining lower temperatures without continuous ice replenishment. See Chiller FAQ and Chiller Max FAQs for more details.
